Line 33 is first designated as a closure line for line 3 during the reconstruction of the tram track between Dolní and Horní Hanychov. This was the case from 9 July 2001, when line 33 replaced line X3 on the same route. Until 1 May 2004 transport on this line was provided by the private operator Ivan Pacák. The need for the line ended with the ceremonial opening of the tram track on this section on 1 August 2005.
Line 33's second appearance begins on 1 September 2006, when it is introduced already on today's route, that is, KUNRATICKÁ SÍDLIŠTĚ - Dobiášova - Rochlice - PRŮMYSLOVÁ ZÓNA JIH. It begins as three services in the outbound direction for shift changes and three services back after the change. These replaced line 35 services on a similar route. The line caught on very well — in 2011 a morning reinforcement service from the U Lomu stop was added, since 2018 an afternoon reinforcement service runs from the Krejčího stop, and most services are operated by articulated buses.
As with the other lines serving the southern industrial zone, with line 33 too it is crucial which service it is. Between Rochlice and Kunratická none of them tends to have a problem, but, for example, the service distributing the morning shift can pick up as much as a twenty-minute delay at the start of the route.
